A sequential double-label autoradiographic 2-deoxyglucose study was performed to investigate a potential role of either the substantia nigra or the ventromedial thalamus in partial onset cortical seizures in the rats. The double-label procedure consisted of three phases of control labeling by 14C-2 DG with recurrent seizures induced by low frequency cortical stimulation (LFCS), electrolytic lesion placement in either the substantia nigra or the ventromedial thalamus, and test labeling by 3H-2 DG under the same experimental condition with the control uptake period. Each tracer image was separately estimated through X-ray film and 3H sensitive film autoradiograms of the same brain slice. Subtraction of the first tracer image from the second tracer image was then accomplished to extract altered metabolic rate specifically related to the lesion placement. The difference image was evaluated statistically after having precisely accounted for the test-retest variability.
