Etiologies of various malformations of the human brain include genetic and environmental factors. In many of the anomalies resulting from single gene mutations, the responsible genes have been identified since the 1990s. Holoprosencephaly is caused by defects of the Sonic hedgehog signaling pathway that modulates the patterning of forebrain, whereas neuronal migration disorders result from defects of molecules that regulate a specific aspect of neuronal migration. By contrast, environmental factors, such as folate deficiency, play a major role in neural tube defects. Studies of these factors have led to the identification of genetic causes such as the polymorphism of an enzyme in folate metabolism.
