It is known that plasma membranes of kidney cortex, aorta, and adrenal cortex possess high affinity specific binding sites for atrial natriuretic poly-peptide (ANP). We characterized ANP-binding prop-erties of plasma membrane fractions obtained from human and rabbit lung. Guanylate cyclase activity of the fraction of human lung was also determined. Scatchard analysis of the binding properties reve-aled the presence of a single high-affinity binding site, Kd =48.5 pM, B max =63.8 fmol/mg protein for rabbit lung and Kd =91.3±51.6 pmol/l, B max= 26.2 ±17.6 fmol/mg protein for human lung (N=3). The human lung plasma membrane fraction produced cyclic GMP (cGMP) in a dose-dependent manner by addition of ANP. Though prominent increases in cGMP were observed at 1-40 nmol/l ANP, the in-creasing trend was observed at 10 pmol/l ANP. These findings suggest that human lung plasma membrane is equipped with specific ANP receptors and that the lung may be one of the target organs for ANP.
