生体内で微量にしか存在せず，半減期も短い生理活性脂質の生理作用は，遺伝子工学の手法を駆使した受容体や産生酵素の研究によってよく理解されるようになってきた．生理活性脂質は細胞膜受容体・核内受容体のいずれかに結合してその生理作用を発揮するが，これまで核内受容体を介してのみ作用すると考えられてきたステロイド由来のいくつかの脂質に対しても細胞膜受容体が同定された．生理活性脂質に対する細胞膜受容体の多くはGタンパク質共役型受容体であり，活性化に伴う構造変化がX線結晶構造解析によって明らかにされた．新規の生理活性脂質・脂溶性リガンドの同定には高感度質量分析機が有効であり，今後の臨床検査への応用が期待される．

Recent molecular biology techniques enabled us to reveal the functions of bioactive lipid mediators and their receptors.　Lipid mediators transduce signals through either membrane receptors or nuclear receptors, and recently certain lipids derived from cholesterol were found to activate both of them.　All the membrane receptors for lipid mediators are G protein-coupled receptors (GPCRs).　Recent crystallographic analysis revealed the dynamic structural change of GPCR upon ligand binding using β2 adrenergic receptor as a model.　Highly sensitive mass spectrometry is useful for the identification of novel bioactive lipids, and will be used in laboratory tests for diagnosis in the near future.
