脳腫瘍におけるproliferating cell nuclear antigen（PCNA）発現の解析

Analysis of proliferating cell nuclear antigen (PCNA) expression in brain tumors

I．はじめに

　脳腫瘍は頭蓋腔という閉鎖された腔内で発育増殖する新生物であり，腫瘍細胞の増殖能，増殖速度の程度が他臓器の腫瘍以上に患者の予後を大きく左右する重要な因子となる．近年，腫瘍細胞の増殖能を反映する種々の増殖細胞核内抗原に対するモノクローナル抗体が開発され^9-11,18），免疫組織学的染色により腫瘍細胞の増殖能の評価が可能となり，病理組織学的所見と併せて脳腫瘍の悪性度や予後の検討に用いられるようになった^{5,13,15,24，26-28,30}．なかでもDNA合成酵素DNA polymeraseδの補助蛋白であるproliferating cell nuclear antigen（PCNA）^4,22）の免疫組織学的検索は，パラフィン包埋切片でも可能であるという利点から最近広く行われている^{1,7,8,12,23,25）}．しかし，抗PCNA抗体を用いた免疫組織学的染色では個々の陽性細胞の染色強度に差がみられ，測定者による陽性細胞率の値が一定しないという問題点をはらんでいる．また，PCNAは細胞周期のG1後期からS期にかけて特異的に発現する^2,6,16,20）と言われているがその詳細については不明である．

　今回われわれは，細胞周期とPCNA発現との関係をC6ラットグリオーマ細胞を用いた同調培養法により検討した．また脳腫瘍におけるPCNAの発現を免疫組織学的染色法及び定量を意図したimmunoblotting法により検索し，その有用性について検討した．

The expression of PCNA in brain tumor cells was measured in vitro and in situ by using an electrophore-tical immunoblotting method and an immunohistoche-mical staining method. In synchronized C6 rat glioma cells, PCNA were almost parallel to the S phase, although the small amounts of PCNA were expressed in G1 and G2 M phases. In astrocytic tumors from op-erative tissues, immunohistochemical PCNA positive rate increased significantly with increasing tumor grade. PCNA positive rate of recurrent meningiomas was also significantly higher than that in nonrecurrent meningiomas. These findings suggest that the PCNA is associated with the cell cycle, especially the S phase and immunohistochemical staining of PCNA is useful to evaluate the proliferating activity of a brain tumor. Immunoblotting method would also be helpful for the exact analysis of proliferating activity in brain tumor cells.
