破骨細胞の分化は,骨芽細胞系細胞から分泌されるreceptor activator of NF-κB ligand, colony stimulating factor-1やosteoprotegerinによって調節されている。骨形成におけるWnt古典経路と非古典経路の役割の解明に伴い,骨吸収におけるWntシグナルの役割も解明されつつある。 本総説では,破骨細胞の分化におけるWntシグナルの役割について概説する。
Osteoclast differentiation is regulated by receptor activator of NF-κB ligand, colony stimulating factor-1 and osteoprotegerin. These cytokines are derived from osteoblast-lineage cells such as osteoblasts and osteocytes. Wnt has been shown to be critical for osteoblast differentiation throughβ-catenin dependent canonical andβ-catenin independent noncanonical Wnt signals. Recent studies have shown that osteoclastogenesis is also regulated by Wnt signals. This review summarizes recent advances in roles of Wnt signals in bone resorption under physiological and pathological conditions.
