Absorption and metabolism of alcohol and genetic polymorphisms of alcohol metabolizing enzymes and their associations with alcohol dependence were described. Alcohol is mainly metabolized by ADH1B and ALDH2 in the liver, but cytochrome P450 and catalase are also involved in alcohol metabolism. In Asians, genetic polymorphisms exist in both ADH1B and ALDH2 and they are associated with onset of alcohol dependence. The inactive form of ALDH2 and super-active form of ADH1B play roles in preventing alcohol dependence independently. However, alcohol dependence is a heterogeneous disorder and many other factors, including environmental factors, relate to the onset of alcohol dependence.
