The role of second messengers (inositol 1,4,5-trisphosphate and diacylglycerol) on membrane conductance was examined in voltageclamped NG108-15 neuroblastoma x glioma hybrid cells. A brief local application of bradykinin (BK) evoked a transient outward current, followed by a sustained inward current. The initial outward current was duplicated by intracellular injections of inositol 1,4,5-trisphosphate. The second inward current was replicated by extracellular perfusion with phorbol 12,13-dibutyrate. The result suggests a dual, time-dependent role for these two intracellular messengers in the modulation of membrane excitability by BK.
