HLAと連鎖した疾患感受性遺伝子の同定およびその環境要因との相互作用

HLA-linked genes controlling the susceptibility to neurological disorders

はじめに

　疾患の発症要因は，遺伝要因と環境要因とに人別される（図1）。遺伝要因のみで発症する疾患は，いわゆる古典的な単因子遺伝病でその遺伝様式もメンデルの法則に従う単純なものである。これらの疾患は，構造遺伝子や遺伝子発現の調節に関与する遺伝情報に生じた変異に起因すると考えられる。環境要因のみで発症する疾患としては，外傷，中毒あるいは多くの急性感染症があげられる。このほかの大多数の疾患は，複数の遺伝要因（polygene）と複数の環境要因を必要とする多因子疾患（multifactorial disease）であり，その遺伝様式（多因子遺伝）も複雑である。われわれは，ヒト第6染色体短腕上に位置するヒト主要組織適合抗原系HLAと連鎖した疾患感受性遺伝子の解析を行なっており，本稿では，このうちより神経疾患の発症を支配する遺伝要因およびその環境要因との相互作用に関して述べたいと思う。

Abstract

Both the genetic and the environmental factors control the development of many diseases. In this review, we have indicated the existence of HLA-linked genes controlling the disease susceptibility to neurological disorders with or without interac-tion of the environmental factors.

Autosomal dominant type of spinocerebellar ataxia (SCA) with spasticity (Marie's ataxia) was linked to HLA and the maximum lod score for the linkage between HLA and SCA was 3.15 at the recombination fraction 0.12. HLA typing of the family members is, therefore, very useful to the prediction and the diagnosis of SCA in the family with SCA.
