異種移植ヒトグリオーマ細胞の電顕観察—とくに細胞分化について

Ultrastructural observation of heterotransplanted human glioma cells—Special reference to cell differentiation

I．はじめに

　未分化中枢神経腫瘍の分化の問題は古くから注目されている^14,15）。これら腫瘍の形態分化の研究から，中枢神経腫瘍の組織像の解釈が容易となり，ニューロンまたはグリア細胞分化に関する知見が豊富となる。理論的には，クローン化された腫瘍細胞が異種組織内に移植され，そこで増殖が可能であれば，それが最良の実験モデルとなろうが，中枢神経腫瘍では，これがきわめて困難である。最近になり，ヌードマウスまたは無脾無胸腺（lasat）マウスを用いた異種移植成功例が報告されるようになった^{1,4,8〜10,18〜21）}。われわれは培養株化ヒトグリオーマ細胞のlasatマウス皮下への移植腫瘍につき検討し，ヒト腫瘍との類似性，組織学的特徴とグリア線維性酸性蛋白陽性所見を報告したが^11,20），ここに移植腫瘍の微細構造を細胞分化を中心に述べる。

Abstract

Ultrastructural observation was done on the tumors heterotransplanted on the back of threehereditary asplenic-athymic (lasat) mice. The explant was an established human glioma cell line designated KNS-42 derived from a human glioblastoma. Two examples presented many small cells with round hyperchromatic nuclei. Ultra-structurally, polysome was the main organelle and the coarse chromatin thread was present evenly in the karyoplasm. The cell surface was usually smooth and did not show any interdigita-tion and special junctional divices.
