小脳形態形成の分子機構

Molecular basis of cerebellar morphogenesls.

小脳の形態形成の過程で働く分子が数多く報告されるようになってきた。初期の神経管の中での前後軸に沿った位置情報の獲得には中脳・後脳境界部のオーガナイザーの役割が注目される。一方，背腹軸に沿った形態形成では，TGFβシグナルなどの神経管の背側化因子が小脳の発生にも重要な役割を果たす。大まかな位置情報が与えられた後，神経細胞サブタイプの分化が起きる。顆粒細胞への分化の過程ではヘリックスループヘリックス型の転写因子が，プルキンエ細胞の成熟の過程では核内レセプター型の転写因子が関与する。一方，比較的後期の発生段階でおきる小脳内での形態形成については，Shhなどの顆粒細胞前駆細胞の増殖に関わるシグナル，Reelinなどの層構造の形成に関わるシグナルなどが働いていることが，明らかになりつつある。

How are the elaborate structures of the mammalian and avian cerebella established ? This fascinating problem has been investigated by many researchers. Recent advances in the genetic manipulation of mice have increased our understanding of the molecular basis of cerebellar morphogenesis. Cerebellar morphogenesis can be examined at each developmental stage. Initially, regional specification occurs in the neural plate. This event defines the cerebellar territory along both the rostrocaudal and the dorsoventral (medial-lateral) axes.
