Botulinum type A toxin has been studied as a treatment of choice for human movement disorders since 1970's. It works at the cholinergic nerve terminal and blocks calcium-mediated acetylcholine release by cleaving cytoplasmic protein associated with the exocytosis of synaptic vesicle, SNAP-25. The resultant neuromuscular block causes muscular weakness and atrophy, as seen in botulism. The activity of crystalline toxin is about 3.0×107 (±20%) mouse intraperitoneal LD50 (units) /mg protein, which is the most potent substance in the nature.
