In the brain, various gene products are induced after epileptic seizure, and they may affect seizure development and seizure-induced neuronal loss. In this review, we overview roles of these gene products in pathophysiology of epilepsy. In particular, we focus on an inducible prostaglandin synthase COX-2 that we have studied. COX-2 facilitated the recurrence of seizure and accelerated the neuronal cell death by increasing PGE2 concentration. Furthermore, we observed that PGE2 was produced mainly in vascular endothelial cells after seizure. Further analysis of gene products induced by seizure may provide a better understanding of the process of epilepsy, as well as the development of new treatments.
