Amyloid β-protein(Aβ)plays a pivotal role in the development of Alzheimer's disease. Aβ is produced from β-amyloid precursor protein(APP)through two successive cleavages by β-secretase and γ-secretase. A substantial fraction of β-secretase, BACE1, is localized to the low-buoyant density membrane(LDM)fraction, which is characterized by abundant cholesterol and sphingolipid. Active γ-secretase components, including presenilin N-and C-terminal fragments, glycosylated nicastrin, APH1, and PEN2, also reside in the LDM fraction. Thus, although only a small fraction of APP is present in the LDM fraction, the membrane microdomains in that fraction may play an important role for Aβ production through regulated enzyme-substrate interaction.
