要約 目的:結膜上皮におけるIL-8 mRNA(IL8)とMUC遺伝子(MUC)発現の検討。対象と方法:アレルギー性結膜炎,アトピー性角結膜炎,春季カタル(VKC群)の患者のimpression cytology検体(検討1),およびlipopolysaccarideまたはヒスタミン刺激培養ヒト結膜上皮細胞(HCE)(検討2)で,IL8,MUC1,MUC5AC,MUC16発現をreal-time RT-PCR法で検討した。結果:検討1:IL8発現は,VKC群で高値を示し(P<0.05),IL8とMUC1は相関した(r=0.54, P<0.05, 偏相関検定)。検討2:ヒスタミン刺激したHCEでIL8とMUC1が相関した(r=0.62, P<0.01, 偏相関検定)。結論:結膜上皮でのIL-8とMUC1の変化は,VKCのアレルギー炎症でみられる。
Abstract. Purpose:To investigate the IL-8 mRNA and MUC gene expression in conjunctival epithelium. Subjects and Methods:Gene expression of IL8, MUC1, MUC5AC, and MUC16 in specimens from patients with allergic conjunctivitis, atopic keratoconjunctivitis and Vernal keratoconjunctivitis(VKC group)using impression cytology(investigation 1), and cultured human conjunctival epithelial cells(HEC)stimulated by lipopolysaccharide or histamine(investigation 2)was determined by real-time RT-PCR method. Results:Investigation 1;IL8 expression in VKC group increased, and correlated with MUC1 expression(r=0.54, p<0.05, partial correlation). Experiment 1;Correlation between IL8 and MUC1 expression was shown in histamine stimulated HCE(r=0.62, p<0.01, partial correlation). Conclusion:Alterations of IL-8 and MUC1 in the conjunctival epithelium are demonstrated for allergic inflammation in VKC.
