The effects of water-soluble ionized (Diatrizoate meglumine/Na, Meglumine iothalamate and Meglu-mine iocarmate) and non-ionized (Metrizamide) con-trast media on the cerebral cortex of the rat were studied and compared with the effects of Ouabain and Penicillin. EEG, somatosensory evoked potential (SEP) were recorded and served as indicator of spontaneous and synaptically evoked cortical ac-tivity.

Ionized contrast media were potent convulsant of Penicillin type. Like Penicillin, they enhanced the late components of SEP (P2 and N2) and could suppress the penetration of spreading depression (SD) induced by KCL.

On the other hand, Metrizamide and Ouabain caused marked non-convulsive disturbances such as depression and slowing of EEG. At the same time, the amplitude of SEP was also depressed, and SD induced by KCL was enhanced and prolonged. Similar pharmacological action patterns were cor-related with similar patterns of ultrastructuralpathological changes. Like Penicillin, Diatrizoate meglumine/Na caused marked swelling of dendrites with rather preserved appearance of presynaptic terminals and synaptic vesicles. Metrizamide and Ouabain induced ultrastructural changes involving dendrites, axon terminal and presynaptic vesicles.

The non-ionized molecular structure may endow Metrizamide with affinity to cell membrane and may cause non-convulsive effects on the cerebral cortex like Ouabain. The clinical findings which are recognized as generalized slowing of EEG fol-lowing the myelography with Metrizamide by some authors may also be explained by these actions of Metrizamide.
