Experimental and clinical studies of chemotherapy of malignant glioma were performed. In this report (part 1), the anti-brain tumor activity in vitro. of bleomycin, BCNU and cyclo-cytidine were verified using the cultured glioma cells.

The influence of the agents on these cells was judged by the growth curve and 3H-thym dine uptake.

Results :

1) These agents inhibited the growth curve of glioma cells (more than 50% of the control) at the concentration of 1-10 mcg/ml.

2) The cells of the faster cell cycle were more inhibited than the cells of slower cell cycle was done.

3) Even in the same cells, the inhibition of the growth rate was different in the different conditions. The cells in the better nutritional condition were more inhibited.

4) The most sensitive agent was different in each cell.

5) The dose response of each agent on the in- hibition of the growth rate was also different in each agents and cells.

6) The agents (BLM, BCNU and cyclo-cytidine) inhibited the 3H-thymidine uptake of these cells at the concentration of 1-10 mcgiml.

7) The degree of the inhibition observed by the 3H-thymidine uptake was almost the same with one seen by the growth curve.

These results indicated that the each cells and agent has its individuality, and DNA metabolism of these cells almost depended on the salvage pathway.

This fact suggested us the necessity of the screen-ing in vitro. of each clinical glioma.
