Morphological and Biochemical Properties of a Long-term Line of Tissue Culture derived from Glialblastoma Multiforme

Ⅰ．緒言

　脳腫瘍の体外培養は，その手技が比較的容易であるにもかかわらず，多くの研究者が携わるようになつたのは1960年以降である。もちろんそれ以前にもFischer^1），Cushing^2），Russel^3）らの古典的論文もあるが，現在の発展を見たのは1961年Kersting^4），1963年Lumsden^5）によるところが大きい。そして，これらの多くの人々の目的とするところは，おそらくこの方法を通じて腫瘍の細胞由来を探り，またこれを扱うことにより生体内の状態を推定し，診断治療法開拓の実験系とすることの二つであつたろう。筆者らは主として後者を目的として培養を始めたが幸い長期継代培養によりmalignant glioma由来細胞の株化に成功した。そしてこれを利用して脳腫瘍化学療法^6），7），放射線療法のための実験^8）が行なわれている。本稿にはその長期継代の経過と現在まで得られたいくつかの生化学的性状を別途に継代したmeningioma細胞，oligodendroglioma細胞と比較して述べる。

A cell line derived from human glioblastoma multiforme was established and still continues vigorous growth at the 37th passage (12 months cultivation).

Morphological and bichemical properties of the cell line were studied with the following results; 1) No remarkable changes of morphology have been observed from the primary culture up to the present 37th passage.

2) In Na⁺ free solution, the cells showed the bubbling formation at 2 min., vacuolization and filamentous mitochondria at 15 min., and destroy of nuclei membrane at 60 min.
