  • Abstract
  • Look Inside

It has been demonstrated in biochemical and pharmacologic studies that in the monoamine-neuropeptide interactions in the hypothalamus, the monoamines play some important roles in regulating the secretion of both neurohypophyseal and hypophyseotropic hormones. Also recently the monoaminergic synaptic inputs to some brain hormone-producing neurons in the hypothalamus have been revealed by electron microscopy. In this study, the monoaminergic synaptic inputs to the neurons containing neurohypophyseal or hypophyseotropic hormones in the rat hypothalamus were examined and described using a combined technique combining 5-hydroxydopamine (5-OHDA) labeling or autoradiography after intracerebroventricular injection of 3H-monoamine with brain peptidergic hormone-immunocytochemistry or double labeling immunocytochemistry combining the preembedding peroxidase-antiperoxidase (PAP) method with the postembedding immunogold staining method at the elctron microscopic level. Antisera to the following brain peptidergic hormones were used to detect them by immunocytochemistry: argininevasopressin (VP), oxytocin (OT), thyrotropin-releasing hormone (TRH), luteinizing hormone-releasing hormone (LHRH), corticotropin-releasing hormone (CRH), somatostatin (SRIF), growth hormone releasing hormone (GRH), S-endorphin, ACTH, dynorphin and neurotensin.

 1. Monoaminergic innervation of neurohypophyseal hormone-producing neurons

Using a technique combining autoradiography with immunocytochemistry, we have found the synapses between 3H-NA or 5-OHDA-labeled axon terminals and VP-like immunoreactive nerve cell bodies and fibers in the paraventricular nucleus (PVN) and supraoptic nucleus (SON) of the rat hypothalamus, and the synapses between 3H-NA- or 5-OHDA-labeled axon terminals and OT-like immunoreactive neurons in the PVN.
