脳のロイコトリエンとその生理的作用

Physiological and pathophysiological role of leukotrienes in the central nervous system.

I．はじめに

　脳は他の臓器と同様にあるいはそれ以上に，活発なアラキドン代謝の部位である。脳におけるアラキドン代謝の研究は，シクロオキシナーゼ経路のプロスタグランディン（PG）を中心に展開され，PGD₂，PGE₂あるいはPGF₂αなどが脳内において生合成され，かつ睡眠，体温，ホルモンの遊離などの調節因子として重要な生理的役割を演じていることが明らかにされてきた^1〜3）。一方最近の研究では，脳にはリポキシゲナーゼ経路も存在し，LTB₄，LTC₄などのロイコトリエン（LT）を生合成する能力を有していることが明らかにされ，LHやLH-RHの遊離調節因子などの生理的役割に加えて，脳浮腫や脳虚血などにおける病理的役割も注目されている。本稿では，脳におけるLTの生合成と脳内分布を中心に，脳におけるLTの生理的および病理的役割について紹介する。

Recent studies have demonstrated the formation of leukotrienes in the central nervous system and have also provided evidences for physiological and pathophysiological role of leukotrienes in the brain. This review is intended to provide the reader with a brief overview of leukotriene biology, with specific reference to the central nervous system. The increasing knowledge about the role of leukotrienes in neurobiology may contribute to the understanding and treatment of some neurological diseases.
