Muscarinic ACh receptors : nature of the bindings of radiolabeled ligands

　アセチルコリンレセプター（ACh-R）を，分子レベルでとらえようと試みた研究が，1960年代中頃より，アイソトープで標識した受容体作働薬（agonist），遮断薬（antagonist）の結合の性質を研究することで始まった。ACh-Rには，ニコチニックとムスカリニックの2種存在することは，よく知られているが，前者のニコチニックレセプターの研究は，α-ブンガロトキシンなどヘビ毒トキシンという非常に親和性の高い結合物質の利用，シビレエイなど発電魚の発電器官という多量にニコチニックレセプターを含む材料，さらにニコチニックレセプターが界画活性剤で失活しないという特性などより，ほぼ単一の蛋白として精製が進み，その分子的性質が明らかになりつつある。

　一方，ムスカリニックレセプターについては，まだ種々のagonists，antagonistsの結合の性質を見ている段階であり，分子レベルでの性質が明らかになったとは言えないが，これまでの研究を通じて，従来のレセプター理論では説明できないような現象が観察されている。

Abstract

　The molecular characters of muscarinic receptors have been studied since 1965 by use of ³H-labeled ligands which bind specifically to muscarinic receptors. In those binding studies, the phenomena, which could not be explained with the classical occupation theory for receptor-ligand interaction, were observed. In this review, we summarize the nature of the bindings of agonist and antagonist to muscarinic receptors and also mention the changes in amount of muscarinic receptor at several conditions mainly according to our results.
