Apoptosis in muscular dystrophy

1995年，私たちはエバンスブルーによる筋変性領域を可視化する生体染色法を開発した。これをアポトーシスを証明するTUNEL法と組み合わせてヒトのデュシェンヌ型筋ジストロフィー（DMD）のモデル動物であるmdxマウスに応用したところ，エバンスブルーの細胞膜透過性が昂進している筋変性領域に限局してアポトーシスが起きていることを見出した。当時は筋ジストロフィーの病態発生におけるアポトーシスの関与は知られていなかったが，その後，ヒトのDMDを含む多くの筋疾患においてアポトーシスが関与していることが報告されてきた。筋疾患におけるアポトーシスの発生機序の解明は種々の阻害剤による治療法の開発につながることが期待される。

In 1995, we had developed a vital staining method to visualize degenerated muscle fibers in mdx mouse. Combining this vital staining, TUNEL technique and gel electrophoresis of genomic DNA in the degenerating muscle, we had shown that degenerating muscle fibers undergo apoptosis in mdx mouse. Since then, many papers showing apoptosis in dystrophic muscle in animal models and human patients appeared. Now, apoptosis has been recognized as one of key steps in pathogenesis of muscular dystrophy. Understandings on apoptotic processes in dystrophic muscle may provide clues for new development of therapeutic strategies.
