 It is now widely accepted that cytokines have pleiotropic actions on the immune system, hematopoietic system, and nervous system. Recently, cytokine networks in the nervous system, the role of cytokines in inflammatory, autoimmune, or even degenerative disorders of the nervous system, and neurotrophic and differentiation effect of cytokines on neural cells attract particular interest. To understand these, here we review cytokine receptors in the nervous system.

 IL-1 receptors are demonstrated using [125I] IL-1α, β-binding on the neurons of granular and molecular layers in the dentate gyrus, but not in the hypothalamus. Immunoautoradiographic studies using anti-Tac antibody demonstrated the α subunit of IL-2 receptors on neuronal cells in the hippocampus and hypothalamus. We have demonstrated the β subunit (AIC-2A and B) of IL-3 receptors on cholinergic neurons in the basal forebrain and septal region by PCR and immunocytochemical staining. We have also demonstrated that IL-3 receptor-associated antigen (IL-3 RAA) is present on widely distributed neurons using F9 monoclonal antibody. Erythropoietin receptors have been demonstrated on PC12 cells as well as central cholinergic hybrid cell line SN6 cells. IL-6 receptor α subunits exist in pyramidal cells and granule cells of the dentate gyrus and hippocampus. It is shown that the receptors for IL-6, leukemia inhibitory factor, and ciliary neurotrophic factor share IL-6 signal transducer gp130. Other receptors for steel factor, colony-stimulating factors, tumor necrosis factor a, interferon α, transforming growth factor β, and activin are also discussed.
