 Bone cancer pain is difficult to treat and has a strong impact on the quality of life of the cancer patients. Now, we have tried to analyze a mouse model of bone cancer pain. To date, we found two mechanisms of bone cancer pain. One is the down-regulation of μ opioid receptor expression and the up-regulation of CCKB receptor in peripheral nervous system. The other is the involvement of TRPV1. TRPV1 expression was increased in the DRGs in a transcription-dependent fashion, and pharmacological blockade of TRPV1 reduced bone cancer-related pain behaviors. In addition, the activation of TRPV1 is required for pain induced by endothelin-1, which is an important mediator for bone cancer pain. Therefore, TRPV1 activation plays a critical role in the generation of bone cancer pain.
