 In the genetic studies of bipolar disorder, genes commonly associated with schizophrenia and bipolar disorder have attracted the researchers. Among them, G72 is most replicated genetic association in bipolar disorder. However, risk allele is not consistent across studies. Other approaches, such as study of syndromal bipolar disorder, analysis of positional candidate genes, identification of candidate genes by gene expression analysis have been performed. Interesting gene environmental interaction between serotonin transporter polymorphism and stress/maltreatment as risk factors for depression was not supported by subsequent large scale studies. Strong association of depression with loss of function mutation in TPH2(tryptophan hydroxylase 2)was not supported by subsequent studies. Causes of these lack of consistency are discussed.
