網膜色素変性におよぼすα—Methyl-Dopaの効果

Effect of Alpha-Methyl-Dopa (Aldomet) on pigmentary degeneration of the retina

I．緒言

　α—Methyl-Dopa （α—Methyl−3,4 Dihydro—xy-Phenylalanine）は，次に示す構造式を有する化合物で，1954年Sourkesにより，Dopa脱炭酸酵素の拮抗阻害剤であることがみいだされた。その薬理作用はまだ全容が明らかではないが，EpinephrineおよびSerotoninの生合成の過程において，Dihydroxyphenylalanineおよび5—Hydroxytryptophanのdecarboxylationを抑制するといわれている。内科的には褐色細胞腫および高血圧症の治療に用いられているが，今回私どもは日本メルク万有株式会社よりα—Me—thyl-Dopa製剤としてAldomet^1）の提供を受け，網膜色素変性患者2例に使用した成績を報告する。

Favourable results in the treatment of pig-mentary degeneration of the retina had been obtained with reserpin (Serpasil Ciba). (Uchi-da, Jap. J.Ophth., Vol.6 No. 2, 1962). Reserpin was said to be a serotonin releaser in the central nervous system, reducing serotonin in the brain.Alphamethyl-dopa (Aldomet) shard with reserpin the serotonin reducing action in the brain.

Assuming that the effectiveness of reserpin in the treatment of the pigmentary degener-ation of the retina was related to the serotonin reducing action in the brain, a favourable result was expected of Aldomet.
