Flecainide acetateのモルモット左心房筋に対する電気生理学的作用を微小ガラス電極法を用いて行った。静止膜電位は用いた濃度(10−7〜3×10−5M)では不変であったが,オーバーシュートは3×10−6M以上の濃度で減少した。有効不応期は10-5Mの濃度にて有意に延長した。1Hz刺激においてVmaxの減少は3×10−6Mの濃度で約25%,10−5Mの濃度で約50%であった。3×10−6Mで標本を灌流するとVmaxの減少率は刺激頻度が0.2Hzでは10%,0.5Hzでは15%,1Hzでは22%,2Hzでは33%,3Hzでは34%であった。Flecainideは心房筋に対しても明らかな濃度および頻度依存性のVmax抑制作用を有し,心房性頻拍に対しても有用性が示唆される。
Using conventional glass microelectrode technique, electrophysiological effects of flecainide acetate on guinea-pig left atrial muscle fibers were examined. Resting membrane potential was not affected by flecainide at any concentrations tested (10-7M~3×10-5M), although overshoot potential was signifi-cantly decreased at the concentration over 3×10-6M. Effective refractory period significantly increased at 10-5M. The reduction of Vmax was about 25% and 50% by 3×10-6M and 10-5M, respectively at 1Hz. At 3×10-6M, Vmax decreased by about 10%, 15%, 22%, 33%, and 34% at 0.2, 0.5, 1, 2, and 3 Hz respectively. Flecainide, decreased Vmax of atrial muscle fibers in a dose and frequency depen-dent manner. It is suggested that flecainide might be effective in the treatment of atrial tachyarrhy-thmias.
