 The process of morphogenesis of human gastric carcinoma from cancerous cells in the mucosa is still not clear. However, some progress have been made in the study of experimental gastric carcinoma. For several years, a pathohistological study of resected stomach of beagles administered with ENNG p.o. has been conducted. The resected stomach of a dog sacrificed after a six months period of administration of ENNG (total dose: 11.7g) contained a Borrmann 3 type advanced cancer early cancers type Ⅰ and Ⅱc lesions. Microscopic examination of the whole stomach revealed numerous minute cancerl esions (both signetring cell ca. and well-differentiated ca.), hence indicating the multiple developmental foci in the experimental carcinoma. Scrutiny of 5 μm thickness sections in sequence revealed“double-layers structure”in a proper gastric gland. The luminal layer was a normal epithelium whilst cells of layer close to the basement membrane demonstrated characteristics similar to signet-ring cells. Silver impregnation revealed that in the section which showed disruption of basement membrane, signet-ring cells were found to infiltrate into the mucosa propria. Electronic microscopical study confirmed the same fine structures in signet-ring cells found in the layer close to the basement membrane and in the mucosa propria. The above findings indicate that signet-ring cell carcinoma might have developed from the”double-layer structure”in the proper astric gland. The microscopic study at 7.5-month interval of a beagle given 3-month ENNG revealed multiple minute foci consisting of only signet-ring cells, hence confirming the “double-layer structure“ as the origin of the morphogenesis of the carcinoma. Recently, similar“double-layer structure”in a signet-ring cell carcinoma of apatient have been identified. It is hoped that the morphogenesis of human gastric carcinoma could be clarified in the near future.
