 Standard procedure of thiocholine method ofKoelle for histochemical demonstration of AChEhas been recently modified so as to make possiblemicroscopic observation of neuronal AChE intracellularly.

 Results so obtained were compared with histological preparations such as for neurofibrils, Nissl, Substance Golgi-apparatus, and with intravital methylene blue method of Ehrlich. With the modifiedthiocholine method, under phase-contrast microscope in particular, the cytological distribution ofcopper thiocholine sulphate resembled closely theNissl pattern. The result obtained with materialgiven high doses of irreversible inhibitor, D.F.P., revealed also an identical distribution withincytoplasm of the enzymic product. Furthermore, application of Brachet's methylgreen-pyronin testas combined with the thiocholine's, disclosed bothRNA and copper thiocholine product being virtually superimposed or overlapped one another. They were, however, seen in part to be localizedapart.

 These findings were considered from currentview and literature available to be further supportto the hypothesis that intracellular or internal(reserve) AChE is synthesized within or at theendoplasmic reticulum.
