海馬における神経活動と遺伝子発現

Neuronal activity and gene expression in the hippocampus

はじめに

　細胞性がん遺伝子c-fosは培養細胞に成長因子を作用させたとき速やかに発現するが，神経性培養細胞のPC 12をアセチルコリンや高カリウムで刺激した場合にも発現することが見出され^15,25），つづいて動物にけいれんを起こすと海馬などの神経細胞にc-fosが強く発現することがわかった^9,10,24）。C-fOSがコードするFos蛋白は転写調節因子として他の遺伝子発現を引き起こすのであるから，神経細胞の活動でc-fosが発現すると，つぎにそれがどのような遺伝子に働き，その結果産生された蛋白によって神経細胞の構造や機能がどのように変化するのかに関心が集まっている。

　一方，重要な脳内分子としてオピオイドペプチド前駆体やチロシン水酸化酵素の遺伝子解析が早くから行われ，それらのmRNAは神経刺激によって増加することが見出された。たとえばラットの頸部交感神経を90分間電気刺激すると48時間後には上頸神経節のチロシン水酸化酵素mRNAは2.3倍に増加する^13）。

Recent progress in molecular neurobiology has revealed that stimulation of nerve cells induces not only electrical or biochemical responses but also expression of many genes in them. Such immediate early genes as c-fos and zif 268 encode transcription regulatory factors and thus trigger expression of other target genes. Although not yet elucidated, these cascade of gene expression are considered the basis for long-term changes in the nervous system, including neuronal plasticity.
