The separation methods of epidermis from the skin were reviewed for its biochemical studies. Heat and chemical treatments were simple and convenient especially for small biopsied human skin. They could not, however, be used to the enzymatic research for acantholysis of epidermis. Several proteolytic enzymes were also examined. If time and temperature of incubation for the skin were carefully selected, the intact epidermal sheet was able to be obtained even for the use of cell culture. We devised the stretching machine for the mechanical separation to use both experimental animal skin and human biopsied skin. Epidermis obtained by this method was completely intact and was best for biochemical or biophysical studies.
