ウイルスベクターによる神経幹細胞・ES細胞への遺伝子導入

Gene transfer to neural stem cells using viral vectors

　細胞内へ効率よく外来遺伝子を導入し発現させる方法として，遺伝子治療用に開発されたウイルスベクターの利用が有力である。神経幹細胞あるいはES細胞への遺伝子導入の目的には，GFPなどのマーカー遺伝子を発現させることにより細胞を選別し未分化細胞の分化・成熟過程を追跡するマーキングと，転写因子や酵素などの遺伝子を導入することにより特定の表現型を持つ細胞を作成する分化誘導とがある。マーキングには，分裂細胞にのみ感染する特徴をもつレトロウイルスベクターが利用されることが多い。移植治療を目的とする場合には，細胞に傷害を与えずに正常な分化過程になるべく近い状態で特定の細胞を誘導する必要があり，主にレンチウイルスベクターが試みられている。

Viral vectors have been developed to deliver therapeutic genes into a variety of cell types for the treatment of innate and acquired diseases. Neural stem cells and embryonic stem (ES) cells are potential sources of cellular substrates for brain repair and represent attractive candidates for gene transfer using viral vectors. Transgene expression in stem cells is a powerful tool not only for studies of the basic mechanisms of cell differentiation, but also for manipulation of cell fate through genetic modification.
