老年痴呆,Alzheimer病の脳の超微形態的研究により,老人斑は変性神経突起の塊り(Cluster),Amyloid細線維,Alzheimer原線維変化,Synapse変性,Glia突起などにより構成されていることが判明した2,3,11,13,15,17,20,24〜28,30,31)。
老人斑の成因については,神経突起などの変性の結果Amyloidが産生されるという考えと23,26,28)),逆にAmyloidの沈着が一次的で神経突起などの変性は二次的なものとする考え方1,9,20,22,23)が対立している。
Neuronal process degenerations and synapse changes in senile brain, particularly those in senile plaques, were classified into several types according to their characteristic ultrastructural features. Pathogenesis of these changes and amyloid fibrils in senile plaques were also discussed.
Senile plaques were composed of localized cluster of deposited amyloid fibrils, various types of distended neuronal processes, changed synapses, microglia, and astrocytic processes.
