 The incorporation of 3H-thymidine into biopsy specimens of the colon and rectum was studied in vitro autoradiography. In normal conditions, the labeling index showed 8.1% and the labeled cells were richer in the lower third of the crypts. The labeling index in the active stage of ulcerative colitis showed 15.3 and the labeled cells were more abundant in the middle third of the crypts, but the index as well as the site of the labeled cells revealed nothing extraordinary in the stage of remission. In regard to polyps, the labeling index of adenomatous polyps, familial polyposis, and hyperplastic polyps (Lane et al.) indicated 8.7%, 14.4% and 9.1%, respectively. The labeled cells in case of adenomatous polyps and familial polyposis were more often seen in the upper third of the crypts, while those of hyperplastic polyps were more prominent in the lower third of the crypts. As to the epithelium in the surface area, the labeling index of adenomatous polyps, familial polyposis, and villous adenoma showed 9.8%, 14.2% and 10.3%, respectively. But since that of hyperplastic polyps and juvenile polyps indicated 0%, there seemed to be different qualities in the polyps of the two groups. In the case of familial polyposis the labeled cells were, as in adenomatous polyps, more numerous in the upper third of the crypts in the areas that were endoscopically normal but histologically had atypical epitheliums. Marked fluctuations were seen in the labeling index of cancer, which ranged from 9.2% to 34% with a mean of 23.8%.
